It happened during their journey back to Konoha after a mission. The enemies, led by the powerful duo of Ginkaku and Kinkaku composed of twenty dangerous Cloud Jounins.

After the death of their two brothers, Hashirama and Tobirama became determined to find ways to stop the useless fighting.
How Old Was Tobirama When He Died?
Tobirama, during the Cloud incident, was already the Second Hokage. He was appointed Hokage after Hashirama’s natural death. During this period, Hashirama must have been 55 years old by the time he died. It means Tobirama became Hokage at age 54. From this period to his demise, the estimate is that three years have passed. Therefore, Tobirama died in battle at age 57.
He was no longer in his prime when he fought the Cloud Jounins. However, knowing that he successfully protected his whole squad, by distracting the enemy shows how powerful he is. Danzo even stated that no one has measured up yet to Tobirama. It implies that as Hokage he is irreplaceable.
How Did Tobirama Resurrect?
Tobirama’s resurrection happened a few times using the Impure World Reincarnation technique. The first one was during Orochimaru’s siege of Konoha. Hiruzen was forced to fight the two former powerful Hokages.
The third Hokage had to use a forbidden technique to stop the two. Both brothers apologized for giving Hiruzen a tough time. While they were being consumed by the Shinigami, Hiruzen apologized back for his shortcomings.
The second is by Impure World Reincarnation technique performed again by Orochimaru. It was under Sasuke Uchiha’s request. Tobirama was resurrected with the other Hokages, Hashirama, Hiruzen, and Minato.
After the conversation with Sasuke, they headed into the battle. Together with the rest of the Shinobu forces, they fought Madara and the Ten Tailed Beast in Fourth Shinobi World War.
After the infinite Tsukuyomi and appearance of the Sage of the Six paths, all the revived Hokages ascended to the Pure Land. As they fade, Tobirama consoles Hashirama to entrust the future to Naruto and Sasuke.
Did Tobirama Hate the Uchiha?
Tobirama is often accused of hating the Uchiha clan. It is due to how he battled with this clan without mercy before establishing the Leaf. He even mortally wounded Madara’s younger brother that resulted in death. He stopped Madara’s ascension to Hokage by proposing a different way of selecting a leader. The proposal led to Hashirama’s election instead.
Tobirama’s opinion on the Uchiha was based on his rational thinking. Tobirama did not exactly hate the Uchihas. He acknowledged that there were also formidable allies from this clan. But he is aware that the Uchiha’s are like ticking timebombs.
To keep them at bay, he established the Konoha Police Force. The purpose was to channel the Sharingan user’s attention to something useful for peace and order. However, some viewed it as marginalizing the clan. It led to further conflicts.
